We present a novel deterministic search technique for finding a set of maximal conductances in multi-compartmental models, and apply this to a model of a striatal medium spiny neuron. The search rapidly converges on a set of maximal conductances by fitting simulation output to current injection data. The search is robust, finds solutions in the presence of a multitude of inward and outward ionic currents, and is extendible to include a subset of the passive membrane parameters. After incorporating synaptic input, the resulting model of the medium spiny neuron provides a good fit to a range of experimental data.
